The DBT-SAHAJ National Facility for Lifetime imaging at RGCB aims to make cutting edge advanced imaging modalities such as lifetime imaging available to RGCB researchers as well as the wider academic and life science industry community. Additionally, the facility supports training in advanced imaging including FRET imaging, lifetime imaging, highthrough-put imaging and Lifetime/FRET sensor cell development. Large panel of lifetime imaging resources and probes were generated by the facility for training, research and drug screening applications. These resources are available from the facility upon request. The facility also supports advanced and specialised imaging services and highthrough-put imaging services using these resources upon request.

Infrastructure

DBT-SAHAJ National Facility for Lifetime imaging houses the following instrumentation.

1. LEICA FALCON SP8 WLL

The FALCON (FAst Lifetime CONtrast) is a fluorescence lifetime imaging microscopy (FLIM) platform fully integrated in the confocal microscope from Leica SP8 WLL that can deliver video-rate FLIM with pixel-by-pixel quantification. The single molecule detector (SMD HyD) provide a high detection efficiency. The system includes pulsed white light laser source 470-670 nm as well as a 405 nm pulsed laser. All laser lines area available for spectral imaging as well as lifetime imaging. STED lasers: 592nm, 660nm are also inbuilt in to the unit for super resolution imaging. Live imaging is possible under e controlled environmental conditions.

2. NIKON A1 CSU SPINNING DISK HIGHTHROUGH-PUT IMAGER CSU W1

The CSU W1 is a field scanning confocal spinning disk systems from Yokogawa Life Sciences, integrated with Nikon’s AI microscope platform, that supports high speed confocal imaging , Ratio imaging , FRET imaging in highthrough-put applications. Omicron laser lines 405, 440, 488, 562 and 640 enables excitation of most fluorophores and fast emission filter wheel is equipped with filters for diverse FRET probes. Andor Dual EMCCD camera support simultaneous two colour imaging of selected dyes and FRET probes for ratio imaging using NIKON NIS Element software.
